
The protein encoded by this gene is a member of the TNFreceptor associated factor (TRAF) protein family. TRAF proteins areassociated with, and mediate signal transduction from members ofthe TNF receptor superfamily. This protein mediates the signalingnot only from the members of the TNF receptor superfamily, but alsofrom the members of the Toll/IL-1 family. Signals from receptorssuch as CD40, TNFSF11/RANCE and IL-1 have been shown to be mediatedby this protein. This protein also interacts with various proteinkinases including IRAK1/IRAK, SRC and PKCzeta, which provides alink between distinct signaling pathways. This protein functions asa signal transducer in the NF-kappaB pathway that activates IkappaBkinase (IKK) in response to proinflammatory cytokines. Theinteraction of this protein with UBE2N/UBC13, and UBE2V1/UEV1A,which are ubiquitin conjugating enzymes catalyzing the formation ofpolyubiquitin chains, has been found to be required for IKKactivation by this protein. A portion of amino acids 314-343 from the human protein was used as the immunogen for this TRAF6 antibody.
